If I understand your question correctly, you are asking should you use an autoresponder such as Awebber or Constant Contact versus sending the emails from, say Outlook.
In my opinion, it is better to use an autoresponder. If you use Outlook, for example, and your list grows, which is what you want, you run the risk of your emails being tagged as spam. ISP's get antsy when they see large quantities of the same email being sent out at the same time. If they think you are spamming, they will shut down your email account, and that could be disastrous for your business.
Using an autoresponder allows you to automate much of the process of managing your email list. You create a form on your website that allows clients to sign up for your e-newsletter, ecourse, etc. and the autoresponder automaticallys sends it out. Autoresponders also have opt out capabilities built in, a must in complying with CANSPAM regulations.
If cost is a consideration, there are free autoresponders that do a good job. One that comes to mind is MailChimp. The free version limits you to 100 subscribers and a sending limit of 6 times/month.
I am a firm believer in using anything I can to automate my business processes, so I would definitely say go with an autoresponder.
